Professor Gao graduated with the highest honor from USTC and received his PhD degree from Stanford University in 2004. After three years of post-doctoral training at Scripps, he started his independent career at Boston College where he remains as Professor of Chemistry. The Gao group is interested in bioorganic chemistry and chemical biology with a special focus on novel design concepts of molecular therapeutics. Professor Gao’s work has been recognized by a NIH MIRA Award, Sanofi iDEA-iTECH Award, Ono Pharma Science Breakthrough Award, a CAPA Distinguished Faculty Award among others. He is also recipient of Boston College’s 2020 Teaching & Mentoring Award. Deeply committed to community service, Professor Gao served as the President of CAPA (Chinese-American Chemistry & Chemical Biology Professors Association) from 2020 to 2023.
讲座摘要/Abstract:
While therapeutic drugs primarily consist of synthetic small molecules and monoclonal antibodies, peptide-based therapeutics are gaining attraction as they can tap into the benefits of both small molecule and antibody drugs. For example, peptide-based therapeutics have shown promise in inhibiting proteins that frustrate small molecules, yet still able to achieve high stability and potential oral availability that challenge antibody therapeutics. The Gao group at Boston College contributes to peptide drug discovery by introducing new chemistries into peptide design and by developing novel display platforms. Specifically, we have been developing chemically “enhanced” phage libraries, which incorporate multicyclic structures as well as designer covalent warheads to allow discovery of covalent inhibitors. This seminar will discuss the novel chemistries recently developed by the Gao group for covalent binding of amines and for cyclizing peptides on phage. Case studies will also be presented to illustrate the scope of applications for the chemically enhanced phage libraries.
