讲座摘要/Abstract:

Cancer is the major threat to public health. The antibody-drug conjugate(ADC) has become one of the hot spots because of its targeted strong tumor killing effect. Enediynes are one family of the most toxic microbial natural products demonstrating great potential as warheads for the development of ADC anticancer drugs. Among them, 10-membered anthraquinone fused (10-AqF) enediynes, exemplified by Tiancimycins (TNMs), are particularly interesting due to the compact and relatively stable structures. However, the acquisition of such drug leads is challenging with extremely low synthetic or microbial fermentation yields, which hampered their subsequent clinical studies. Recent biosynthetic studies have revealed that 10-AqF enediynes follow a common pathway to achieve their microbial production. Herein, The Zhu lab started from the original TNM-producing strain Streptomyces sp. CB03234, and stepwisely improved the titer of TNMs through strain breeding, medium optimization and multi-omics directed metabolic engineering. Based on these efforts, a practical platform for efficient microbial production of 10-AqF enediynes is constructed that will facilitate their studies in clinical trials.
